[ES] Los objetivos de este trabajo son el diseño, producción, caracterización y aplicación de soportes de cultivo celular inteligentes que respondan con la generación de un campo eléctrico y/o acción mecánica cuando se aplica a distancia un campo magnético externo. Estas membranas estarán compuestas por un compuesto de fluoruro de polivinilideno (PVDF) y nanopartículas magnéticas. PVDF es el polímero con el coeficiente piezoeléctrico más alto cuando cristaliza en la fase beta electroactiva. Mediante el acoplamiento de la piezoelectricidad del PVDF con la respuesta magnética de la nanopartícula, se logra la conversión de un estímulo magnético en uno eléctrico, este se transmite a las células cultivadas sobre la superficie del compuesto, posibilitando así su aplicación como soporte magnetoeléctrico.
La síntesis de las membranas se hará por el método de separación de fases inducida por no solventes (NIPS), una técnica sencilla que garantiza la cristalización del PVDF en estructuras que maximizan su respuesta electroactiva. Se caracterizará la cristalinidad, fase cristalina y microestructura de la membrana. Como un intento de contrarrestar la alta porosidad de las membranas y mejorar la respuesta de las células madre mesenquimales de la médula ósea humana, las hbMSC, se procesarán y recubrirán con una cerámica bioactiva capaz de inducir la formación superficial de cristales de hidroxiapatita. El posprocesamiento de recubrimiento se optimizará para mejorar la aplicabilidad de las membranas sin afectar negativamente el comportamiento electroactivo de los polímeros. Se caracterizarán las membranas en cuanto a sus propiedades químicas, térmicas y morfológicas, así como se evaluará su actividad biológica. Para ello se realizarán ensayos FTIR, DSC, TGA y FE-SEM, así como ensayos para la detección y cuantificación de recubrimientos funcionales. También se realizarán cultivos celulares para evaluar la viabilidad y proliferación de las MSCs.

[EN] The objectives of this work are the design, production, characterization and application of intelligent cell culture supports that respond with the generation of an electric field and/or mechanical action when an external magnetic field is applied at a distance. These membranes will consist of a compound of polyvinylidene fluoride (PVDF) and magnetic nanoparticles. PVDF is the polymer with the highest piezoelectric coefficient when crystallized in the electroactive beta phase. Through coupling of PVDF¿s piezoelectricity with the nanoparticle¿s magnetic response, the conversion of a magnetic stimulus into an electrical one is achieved, this is transmitted to the cells cultured onto the compounds surface, thus enabling its application as a magnetoelectric support.
The supports will be produced using the non-solvent induced phase separation (NIPS) method, a simple technique that ensures the crystallization of PVDF into structures that maximize its electroactive response. The crystallinity, crystalline phase and microstructure of the membrane will be characterized. As an attempt to countervail the membranes high porosity and to improve the response of human bone marrow mesenchymal stem cells, hbMSCs, they will be post-processed and coated with a bioactive ceramic capable of inducing the surficial formation of hydroxyapatite crystals. The coating post-processing will be optimized so that the membranes applicability is improved without negatively affecting the polymers electroactive behaviour. Membranes will be characterized regarding their chemical, thermal and morphological properties as well as evaluated for their biological activity. To this end, FTIR, DSC, TGA and FE-SEM tests will be carried out, as well as assays for the detection and quantification of functional coatings. Cell culturing will also be carried out to assess the viability and proliferation of the MSCs.
