[EN] The management of by-products and agri-food waste is a problem for farmers and agri-food companies. It is necessary to advance in the knowledge of which compounds can be recovered, from which waste, and how to do it so that recovery is sustainable from an economic and environmental point of view. The biotechnological processing of biomass waste, with different microorganisms, allows the production of bioplastics such as polyhydroxyalkanoates (PHAs), usable in several industries for different applications can contribute to reducing the environmental impact associated with the use and accumulation of synthetic plastics.
In this work, the revaluation of rice straw is proposed by obtaining hydrolysates for the culture of Haloferax mediterranei. The subcritical extraction residue with and without bleaching and the rice straw without pre-treatment were subjected to enzymatic hydrolysis with cellulases. The obtention of the different residue samples was successful, together with their hydrolysis, where some parameters regarding enzyme accessibility should be taken into account, and can be perfected for future experiments. The concentration of reducing sugars in the hydrolysate was quantified to identify its potential as a culture medium for the PHA-producing archaea. Although the dinitrosalicylic acid (DNS) assay had to be discarded, the HPLC rendered good results in the determination of the best hydrolysate for biopolymer production. The sample chosen was the residue obtained from subcritical water extraction (SWE) under consideration of the hydrolysate's glucose concentration, saccharification level, and environmental friendliness. Haloferax mediterranei growth and PHA production were good for a first approach, although the optimization of the processes must continue, adjusting parameters and doing more tests.
This bachelor¿s thesis is fully aligned with several of the Sustainable Development Goals (SDG), such as good health and well-being (SDG 3), clean water and sanitation (SDG 6), decent work and economic growth (SDG 8), industry, innovation, and infrastructure (SDG 9), responsible consumption and production (SDG 12), underwater life (SDG 14) and life on land (SDG 15).
