dc.description.abstract | [ES] El gen de la proteína “Ankyrin repeat and kinase domain containing I” (ANKK1) ha sido ampliamente asociado con la vulnerabilidad a trastornos psiquiátricos en los que hay un déficit subyacente del sistema dopaminérgico, a rasgos cognitivos y al aprendizaje. Sin embargo, se desconoce la función de la proteína ANKK1. Las investigaciones realizadas por nuestro grupo han sugerido la relación de ANKK1 con el sistema dopaminérgico y su participación en procesos del neurodesarrollo. Resultados experimentales con técnicas de co-immunoprecipitación y análisis por espectrometría de masas han mostrado que ANKK1 podría interaccionar con proteínas que contienen dominios característicos de factores de intercambio de nucleótidos de guanina y de proteínas que unen el citoesqueleto a la membrana plasmática en las neuronas. El objetivo del presente trabajo es la validación de estas interacciones mediante coimmunoprecipitación específica, estudios de sobreexpresión génica y análisis bioquímico mediante western blot e inmunofluorescencia en células. Se pretende que la validación de estas interacciones sea un primer paso en el descubrimiento de los aspectos mecanísticos y posibles vías fisiológicas en las que participa ANKK1. | es_ES |
dc.description.abstract | [EN] The protein of the gene "Ankyrin repeat and kinase domain containing I" (ANKK1) has been widely associated to vulnerability in psychiatric disorders in which there is an underlying deficit of the dopaminergic system, to cognitive traits and learning. However, the function of the protein ANKK1 is unknown. Previous research by our group has suggested the relationship of ANKK1 with the dopaminergic system and its participation in neurodevelopmental processes. Experimental results of coimmunoprecipitation techniques and mass spectrometry analysis showed that ANKK1 could interact with proteins containing characteristic guanine nucleotide exchange factors domains and with proteins that bind the plasma membrane with the cytoskeleton in neurons. The aim of this study is to validate these interactions by specific co-immunoprecipitation, gene overexpression studies and biochemical analysis by western blot and immunofluorescence in cells. It is intended that the validation of these interactions is the first step in uncovering the mechanistic and possible physiological pathways in which ANKK1 is involved. | es_ES |
