dc.description.abstract | [ES] El género Cucumis incluye especies de gran interés comercial como el melón (C. melo) o el pepino (C. sativus). En la mejora genética de estas especies, el empleo de caracteres de interés agronómico presentes en especies silvestres del género es difícil debido a la existencia de barreras reproductivas interespecíficas (BRIs). El estudio de estas barreras es importante para poder en un futuro determinar los genes responsables e intentar intervenir en su expresión. El objetivo principal de este trabajo es la caracterización de las BRIs en el género Cucumis (Cucurbitaceae). Los objetivos específicos son evaluar las BRIs a nivel pre y postcigótico en un grupo formado por 5 especies silvestres africanas y C. melo, y estudiar la posible correlación entre las BRIs identificadas y las relaciones filogenéticas basadas en el análisis del ADN en 13 especies del género Cucumis. Para la caracterización de las barreras pre- y postcigóticas, se han cruzado las especies dos a dos en invernadero, se ha visualizado el crecimiento de los tubos polínicos mediante microscopía de fluorescencia, se ha determinado el nivel de cuajado y la viabilidad de las semillas obtenidas en los cruces interespecíficos, y se han evaluado los híbridos interspecíficos obtenidos empleando caracteres morfológicos diagnósticos. En base a esta caracterización, se ha confeccionado una escala de afinidad reproductiva entre especies. Esta afinidad reproductiva se comparó con la divergencia genética entre especies empleando polimorfismos de secuencia de ADN obtenidos con GBS (Genotyping by Sequencing), detectando una correlación positiva moderada. | es_ES |
dc.description.abstract | [EN] The genus Cucumis includes species of commercial interest such as melon (C. melo) or cucumber (C. sativus). Breeding of melon and cucumber by using those characters of agronomic interest present in Cucumis wild species is prevented by the presence of Interspecific Reproductive Barriers (IRBs). The study of these barriers is important to identify the genes involved in their expression what may facilitate future modifications of the IRBs. The main objective of this work is the characterization of IRBs in the genus Cucumis (Cucurbitaceae). The specific objectives are to assess the pre- and postzygotic IRBs among 5 African wild species and C. melo, and to study the possible correlation between the identified IRBs and the phylogenetic relationships based on the analysis of DNA in 13 Cucumis species. For the characterization of the pre- and postzygotic barriers, the species have been crossed two by two in a greenhouse, the growth of the pollen tubes has been assessed by fluorescence microscopy, fruit set and seed viability have been measured and the obtained interspecific hybrids have been evaluated by using diagnostic morphological characters. Based on this characterization, a reproductive affinity scale between species has been established. This reproductive affinity has been compared with the genetic divergence between species using DNA sequence polymorphisms obtained with GBS (Genotyping by Sequencing), and a moderate positive correlation was found. | es_ES |
