dc.description.abstract | [ES] El síndrome de Dravet (SD) es una enfermedad rara infantil que consiste en un tipo de epilepsia caracterizada por una variedad de crisis resistentes a fármacos, frecuentemente inducidas por fiebre, que debuta a los pocos meses de edad y que generalmente deriva en discapacidad motora y cognitiva. Esta enfermedad es debida a mutaciones dominantes en el gen SCN1A humano que codifica un canal de sodio activado por voltaje esencial para la excitabilidad neuronal. Existe mucha variabilidad en el fenotipo de la enfermedad, es decir, no hay una sintomatología consenso descrita para todos los pacientes, ni para los que tienen la misma mutación. Para este trabajo se ha empleado el modelo parabss1 de Drosophila melanogaster por ser para el gen homólogo al gen SCN1A en la mosca de la fruta y bss1 una mutación característica de D. melanogaster. Con el fin de intentar entender esta variabilidad presente en la enfermedad, se han testado una serie de genes candidatos a modificadores genéticos del gen para en este modelo. Los modificadores genéticos son genes que no están directamente implicados en la enfermedad, pero alteran o influyen en la expresión de la función de otro gen, segregándose independientemente de la mutación principal. Dichos genes están relacionados con procesos de neurodesarrollo o comunicación sináptica, y tienen su homólogo correspondiente en D. melanogaster. La estrategia genética empleada para alterar su expresión se basa en el uso del sistema UAS-GAL4 para expresar un RNA interferente. La metodología para el estudio de estos once genes es la realización de ensayos de crisis, ensayos de supervivencia y ensayos de locomoción. En los ensayos se ha comprobado la relación de estos 11 genes, candidatos a modificadores genéticos, con el gen mutado parabss1 en el fenotipo de la enfermedad. En el silenciamiento de los genes CG8916, Caα1T, KCNQ, nAChRα4, Clc-a y Slo se observan diferencias importantes en los tres ensayos realizados: disminución del tiempo de recuperación tras una crisis, aumento de la supervivencia y aumento de la longitud recorrida durante un tiempo de exploración, revelando así el silenciamiento de estos genes como supresor del fenotipo del SD. Toy es el único gen modificador en el cual su silenciamiento es potenciador del fenotipo del SD, mostrando una gran disminución de la vida media de las moscas. Estos genes podrían ejercer como dianas terapéuticas y herramientas de diagnóstico del SD. | es_ES |
dc.description.abstract | [EN] Dravet syndrome (DS) is a rare childhood disease consisting of a type of epilepsy characterized by a variety of seizures resistant to drugs, frequently induced by fever. The disease begins at a few months of age and usually results in motor and cognitive disability. This disease is due to dominant mutations in the human SCN1A gene that encodes a voltage-activated sodium channel essential for neuronal excitability. There is a lot of variability in the phenotype of the disease, that is, there is no consensus symptomatology described for all patients, nor for those with the same mutation. For this study, the Drosophila melanogaster parabss1 model has been used because para is the gene homologous to the SCN1A gene in the fruit fly and bss1 is a characteristic mutation of D. melanogaster. In order to try to understand this variability present in the disease, some genes that are candidates for genetic modifiers of the gene para have been tested in this model. Genetic modifiers are genes that are not directly involved in the disease, but alter or influence the expression of the function of another gene, segregating independently of the main mutation. These genes are related to processes of neurodevelopment or synaptic communication, and they have their corresponding counterpart in D. melanogaster. The genetic strategy used to alter its expression is based on the use of the UAS-GAL4 system to express an interfering RNA. The methodology for the study of these genes is the execution of crisis trials, survival trials and locomotion trials. In the trials, the connection of these modifying genes with the mutated parabss1 gene in the disease phenotype has been verified. In the silencing of CG8916, Caα1T, KCNQ, nAChRα4, Clc-a and Slo genes, important differences have been observed in the three tests performed: decreased recovery time after a crisis, increased survival and increase in the length traveled during an exploration time, thus revealing the silencing of these genes as a suppressor of the SD phenotype. Toy is the only modifier gene in which its silencing is an enhancer of the SD phenotype, showing a large decrease in the half-life of flies. These genes could act as therapeutic targets and diagnostic tools for DS. | es_ES |
