dc.description.abstract | [ES] En este trabajo de fin de grado se han sintetizado nanopartículas esféricas de oro (AuNPs) con un diámetro de 2-4 nm, las cuales han sido funcionalizadas con polietilenglicol (PEG), diferentes proteínas (HSA, BSA, RatSA y RabSA) y con ambos isómeros del éster etílico del flurbiprofeno (FBPet). Mediante técnicas como la espectrofotometría UV-vis, la fluorescencia y la fotólisis de destello láser, se ha estudiado la fotorreactividad del complejo FBPet@proteína en distintos medios (acuoso, con AuNPs en disolución y con proteína unida covalentemente a las AuNPs) para poder comparar su comportamiento. Finalmente, se ha realizado un estudio de viabilidad celular con células cancerosas de mama humana (SKBr-3) con el fin de investigar su potencial uso en terapia u otras aplicaciones biológicas. Para la síntesis de las nanopartículas y su funcionalización, se tomaron las cantidades óptimas para llevar a cabo la reacción, siguiéndose el método de Brust-Schiffrin. Mediante las medidas de los espectros de absorción UV-vis, se comprobó que las diferentes proteínas se encontraban correctamente ancladas a las nanopartículas. Tras llevar a cabo el estudio de la fotorreactividad de los complejos en los distintos medios, se obtuvieron diferentes resultados en función de la proteína estudiada. En este sentido, los ensayos realizados con FBPet@HSA, muestran una desactivación de la fluorescencia en presencia de AuNPs, cuya causa puede atribuirse a un proceso de transferencia electrónica entre la nanopartícula y FBPet@HSA, aunque tampoco se puede descartar la formación de un complejo en el estado excitado que no emita. Por último, se realizaron estudios de viabilidad celular con células de cáncer de mama humana SKBr-3. Los resultados obtenidos indican que la tasa de supervivencia de las células es menor en presencia de AuNPs, tanto en disolución como unidas covalentemente a HSA. Asimismo, se ha observado que existe cierta enantiodiferenciación, donde (S)-FBPet parece que induce mayor muerte celular. No obstante, el ensayo debería repetirse con el fin de comprobar la reproducibilidad de los resultados. | es_ES |
dc.description.abstract | [EN] In this work, spherical gold nanoparticles (AuNPs) with a diameter of 2-4 nm have been synthesized and functionalized with polyethylene glycol (PEG), in addition with different proteins (HSA, BSA, RatSA and RabSA); besides, the two isomers of flurbiprofen ethyl ester (FBPet) bound to the proteins have also been used. Spectroscopic techniques such as UV-vis, fluorescence and laser flash photolysis have been used to investigate the photoreactivity of the FBPet@protein complexesin different media (aqueoussolution, with AuNPs in solution and with the protein covalently linked to the surface of AuNPs) in order to compare their behavior. Finally, a cell viability study has been performed with human breast cancer cells (SKBr-3) in order to investigate their potential use in therapy or other biological applications. For the synthesis of the nanoparticles and their functionalization, optimal amounts were taken to carry out the reaction, following the Brust-Schiffrin method. UV-vis absorption spectroscopy was used to confirm that the different proteins were properly bounded to the nanoparticles. After studying the photoreactivity of the complexes in the different media, different results were obtained depending on the investigated protein. In this sense, a significant fluorescence quenching of FBPet@HSA in the presence of AuNPs has been observed, which can be attributed to an electron transfer process between the nanoparticle and FBPet@HSA, although it cannot be discarded the formation of a complex in the excited state with no emission. Finally, cell viability studies were performed with SKBr-3 human breast cancer cells. The obtained results indicate that the cell survival rate is lower in the presence of AuNPs, both in solution and covalently bound to HSA. Also, a slight enantiodifferentiation has been observed, where (S)-FBPet appears to induce greater cell death. However, the assay should be repeated in order to check the reproducibility of the results. | es_ES |
