Phase II study of Pazopanib to evaluate the activity and tolerability in patients with advanced and/or metastatic liposarcoma who have relapsed following therapies or for whom no standard therapy exists

Abstract

[EN] The aim of this study is to evaluate the influence of the angiogenic status of a mixoid sarcoma on the response to Pazopanib. To this effect, three translational phase II studies were performed: the first of them consisted in the evaluation of the serum levels of some angiogenic factors/cytokines at three levels: baseline, after three weeks of treatment, at progression and at maximum response; the second one was based on a PI3KCA mutations analysis so as to demonstrate if those liposarcomas with PI3KCA mutations define a subgroup of patients with differential response to Pazopanib; and finally, we performed a IHQ expression study in order to evaluate microvessel density(MDV) and some genes pathways as well as their role as predictive factors to treatment with Pazopanib.

[ES] El objetivo de este estudio es evaluar la influencia del estado angiogénico de un liposarcoma mixoide en respuesta a Pazopanib. Para ello, tres estudios translacionales fase II fueron llevados a cabo: el primero consistía en la evaluación de los niveles de suero de algunos factores angiogénicos/citoquinas a tres niveles: basal, después de tres semanas de tratamiento, en progresión y en su máxima respuesta; el segundo se basaba en un análisis de mutaciones PI3KCA para demostrar si los liposarcomas con mutaciones en PI3KCA definían un subgrupo de pacientes con respuesta diferencial a Pazopanib; y finalmente, hicimos un estudio de expresión IHQ para evaluar la microdensidad vascular(MDV) y las rutas de algunos genes, así como su función como factores predictivos del tratamiento con Pazopanib.
