[ES] Los TALEs (transcription-activator like effectors) de Xanthomonas funcionan como factores de
transcripción eucariotas en las células vegetales, jugando un papel fundamental en el
desarrollo de enfermedades bacterianas mediante la inducción de genes de susceptibilidad del
huésped. Los TALEs son translocados al núcleo de la célula vegetal y se unen a regiones
promotoras de genes diana de las plantas, cuya transcripción facilita la colonización y
propagación bacteriana. A lo largo de la evolución, algunas plantas se han aprovechado del
mecanismo de funcionamiento de los TALEs para activar genes de resistencia, por medio de
trampas de genes que contienen sitios de unión de efector en sus regiones promotoras, y cuya
activación por el TALE correspondiente desencadena una respuesta de resistencia en el
huésped. La variante AT de Xanthomonas citri ssp. citri desencadena una respuesta
hipersensible hospedante específica (HR) que suprime el desarrollo de la cancrosis de los
cítricos. El efector bacteriano que desencadena este proceso es un TALE de 7.5 repeticiones
llamado PthA4AT. Sin embargo, debido a la corta longitud de este efector y al hecho de
presentar dos repeticiones con reconocimiento ambiguo por dos o más bases, la secuencia
diana de ADN tiene únicamente 8 pb y está presente muchas veces en el genoma de la planta.
La gran cantidad de posibles dianas indica que es necesaria una comprensión más detallada del
modo de acción de PthA4AT, para identificar con éxito la causa de la inducción de la HR.
El objetivo principal de este TFG era evaluar la especificidad de este TALE para desencadenar la
HR en N. benthamiana, con el fin de iniciar la búsqueda de genes diana responsables del
disparo de la respuesta hipersensible. Utilizando un kit de construcción modular de biología
sintética para TALEs, se elaboraron diferentes construcciones derivadas de PthA4AT, se
evaluaron para la HR en la planta modelo Nicotiana benthamiana y se compararon con el
PthA4AT natural, para identificar la especificidad de secuencia requerida por este efector para
disparar la respuesta hipersensible. Los ensayos con las construcciones sintéticas planteadas
en este trabajo han permitido ampliar el conocimiento acerca de la secuencia del gen diana
responsable de la HR en el genoma de N. benthamiana, consiguiéndose reducir el número de
bases posibles para dos de las posiciones del sitio de reconocimiento (la segunda y la cuarta).
Del mismo modo, se llevó a cabo la extensión de la misma en dos posiciones (posiciones
novena y décima del sitio de reconocimiento en el gen diana). Esto ha permitido acotar el
número de posibles genes que podrían ser causantes del disparo de la HR tras la entrada en la
célula de este efector bacteriano.

[EN] Transcription-activator like effectors (TALEs) of Xanthomonas function as eukaryotic
transcription factors in plant cells, playing a central role in promoting bacterial disease by the
induction of host susceptibility genes. TALEs are translocated into the plant cell nucleus and
bind to the promoter regions of plant target genes whose transcription facilitate bacterial
colonization and spread. Over the course of evolution, some plants have co-opted the TALE
mechanism of gene activation for resistance, by means of gene traps that contain effectorbinding-sites in their promoter regions, and whose activation by the corresponding TALE
triggers a host resistance response. Xanthomonas citri ssp. citri variant AT
triggers a hostspecific hypersensitive response (HR) that suppresses citrus canker development. The bacterial
effector that elicits this process is a 7.5-repeats TALE called PthA4AT. However, due to the low
length of this effector, the target DNA sequence is only 8bp long and it is present many times
within the plant genome. The large number of putative targets indicates that a more detailed
understanding of the mode of action of PthA4AT is needed to successfully identify the cause for
the induction of HR.
The main objective of this TFG was to evaluate the specificity of this TALE to trigger HR in N.
benthamiana, to initiate the search of target(s) gene(s) responsible for triggering the HR
response. Using a synthetic biology modular construction kit for TALEs, different artificial
PthA4AT derivative constructs were built and assayed for HR in the model plant Nicotiana
benthamiana, and compared to the natural PthA4AT, in order to identify the sequence
specificity required by this effector for triggering the HR. The trials with the synthetic
constructs raised in this work have allowed expanding the knowledge about the sequence of
the target gene responsible for the HR in the genome of N. benthamiana, being able to reduce
the number of possible bases for two of the positions of the recognition site (the second and
the fourth). In the same way, the extension of the same in two positions (positions ninth and
tenth of the recognition site in the target gene) was carried out. This has allowed us to enclose
the number of possible genes that could be the cause of the HR triggering after the entry into
the cell of this bacterial effector.
